FastPfu FLY DNA Polymerase
TransStart FastPfu Fly DNA Polymerase is a hotstart, ultra-high fidelity and high-processivity DNA Polymerase. It consists in a modified DNA Polymerase, increasing its affinity for dsDNA and enhancing processivity and other caracteristics. Its fidelity is 108 times higher than EasyTaq DNA polymerase making it the DNA Polymerase with the highest fidelity on the market. In addition, its extension rate reaches 6 kb/min. Compared with FastPfu DNA Polymerase, The FLY version of FastPfu DNA High-Fidelity Polymerase has a faster extension rate, higher fidelity, and greater amplification efficiency over regular FastPfu, making FastPfu FLY the best performing DNA polymerase for ultra low non-specific amplification from complex and unpurified gDNA samples, as well as other difficult PCRs targeting GC-rich or AT-rich DNA sequences. FastPfu FLY is available for NGS library prep amplification.

Salut Simon,
La FLY… elle fly en Taqarnac!
Et les colonnes gel/pcr purification fonctionnent très bien.
Tu auras aussi officiellement des acheteurs pour tes colonnes pour 2 labos (ils attendaientt les résultats de mes séquençages pour passer une commande de colonnes chez vous)

In my own hands, you can pick up more targets if you use 68 as the extension temperature.
This will favor the amplification of less stable molecules including AT rich sequences.
Note that this may disfavor amplification of gc rich targets.
If used with PCR stimulant, 68 behave similarly to 72 without PCR stimulant.
I use 68 in 95% of my reactions
If GC rich and very stubborn. Try 75 for extension.

Merci pour tes protocoles. J’ai utilisé le Fast & Steep PCR à quelques reprises et je suis vraiment stupéfait des résultats obtenus en si peu de temps de PCR avec la FLY!

Clonage Fast and Clean. Gel d’agarose pour gel extraction du Vecteur digéré et des fragment Left et Right avant le clonage par assembly CU101. Go Fly!
